Background:Pharmacists have the responsibility to assess the safety and efficacy of any prescription medication. The aim of this study was to develop a validated pharmacokinetic (PK) analysis (AOM) of buspar for treatment of anxiety and depression. Materials and methods: This was a cross-sectional, descriptive, multi-stage, parallel-group, single-dose, pharmacokinetic analysis (PKA) study. A 12-week pharmacokinetic (PK) study was conducted in a non-responder group (n = 20) and a healthy control group (n = 20). In the study, the patients who met the inclusion criteria were enrolled. The study was conducted according to the ethical principles of the Declaration of Helsinki and the protocol was approved by the Ethics Committee of the School of Pharmacy, University of the Western Cape. The study was conducted in accordance with the approved protocol. All patients had a thorough medical history including: a previous history of psychiatric disorders, current use of monoamine oxidase inhibitors (MAOIs) or MAOIs with a sedating effect or with a severe history of depression. The patients had been treated with either benzodiazepines or selective serotonin re-uptake inhibitors (SSRIs), and they had been taking benzodiazepines for a long time. The patients had a positive family history of psychiatric disorders, taking monoamine oxidase inhibitors, or SSRIs, or had a positive family history of psychiatric disorders. The patients were not taking monoamine oxidase inhibitors. The patients were treated with buspar for anxiety and depression. The patients were randomized to the treatment group and the control group. They were enrolled in the study at baseline, 3, 6, and 12 weeks after the last buspar dose. The patients were followed at 3, 6, and 12 weeks. The study was approved by the Ethics Committee of the School of Pharmacy, University of the Western Cape. The patients were enrolled and monitored during the treatment with buspar. The patients were followed at each visit, and the data were analyzed by a validated pharmacokinetic analysis (AOM) analysis. AOM analysis was performed for the study period.
Table 1: Patient characteristics, weight, age, concomitant medications, baseline AOM AOR, baseline PK parameters and AOM parameters. P values were calculated by the independent-sample t-test.The study had several ethical limitations:a short patient population of the patients, which may affect the results; a high percentage of patients in the active control group who have had an active seizure (defined as an episode of seizure activity within 12 weeks); and a lack of adequate control of the patients' anxiety. Patients in the study group had a higher incidence of depression than the control group. AOM analysis could not be carried out to evaluate the efficacy of buspar for anxiety and depression. AOM analysis was performed by means of the pharmacokinetic analysis, and there were no other important variables in the study. The study had some limitations, such as: the patients were selected from the general population and the study group was small; there was a possible risk of bias in the study and the study was not carried out at the time of the data collection. This study was conducted within the limitations of the ethics committee. The study had a number of limitations that should be considered. The results of this study may not be generalizable to other population groups or to the wider population. A larger, multi-stage, parallel-group, pharmacokinetic analysis study will be required to provide more information regarding the safety and efficacy of buspar.
Table 2: Safety and efficacy of buspar for anxiety and depression.Table 3: Safety and efficacy of buspar for anxiety and depression. The PK results showed that the buspar dose was not associated with changes in the plasma concentrations of buspar (Cmax, 1.4 mg/kg), but buspar did have a small effect on the concentration of buspar. The Cmax and the AUC values were not significantly different. However, it should be noted that the PK parameters were not reported in this study. The PK parameters in the buspar treatment group were not significantly different than the control group. The AOM analysis revealed that buspar treatment was well tolerated. The most commonly reported side effects were headache, drowsiness, and sedation.
